Induction Shoot At Culture Callus Potato (Solanum tuberosum L.) With Various Concentrations Of Coconut Milk In Vitro)

The importance of the used of superior quality seeds and expected to increase potato production. Potato farmers do not currently used a lot of quality seed potatoes for potato tuber prices are relatively expensive and difficult resulted. The aimed of this research was to determine the concentration of the used of coconut milk with direct pipetted technique is right in stimulated callus induction in potato shoots in vitro culture techniques. This research was conducted in October s.d January 2017 in tissue culture laboratory State Polytechnic of Jember. The experimental design that was used a completely randomized design (CRD) with 2 factors. The first factor treatment of different concentrations of coconut milk are 8 level of concentration of coconut milk, which is 10 ml, 20 ml , 30 ml, 40 ml, 50 ml, 60 ml, 70 ml and 80 ml of coconut milk. The second factor is the concentration difference of 2,4-D on the source callus consisted of 9.05 μM and 13.58 μM. The parameters measured include the time appeared Roots, the number of roots, root length, appeared of shoots, the number of shoots and the long shoots of potato plantlets. while other data were analyzed by analysis of variance (ANOVA). Results of combination treatment research granted coconut milk and 2 levels callus cultures showed non significant response (ns) on the observation parameters measured include the time appeared roots , the number of roots, root length, appeared of shoots, the number of shoots and the long shoots of potato plantlets. While the parameters measured include the time appeared roots, factor L (source callus) provides highly significant results (**). The Faktor L did to LSD 5% which the result of L1 (1,53b) The better from L2 (2,37a).
