ABSTRACT
This research was conducted to determine the concentration of variations of auxin 2.4-D growth regulators that are appropriate for inducing sugarcane callus PS 862. The implementation of this research was held at the State Polytechnic of Jember specifically at Tissue Culture Laboratory from September 26th 2017 to November 23rd 2017. The experimental design was used is a Completely Randomized Design (CRD) using a single factor with 4 replications and 6 treatments namely A1 = 0 ppm Auxin with 2,4-D, A2 = 1 ppm Auxin with 2,4-D A3 = 2 ppm Auxin with 2,4-D, A4 = 3 ppm Auxin with 2,4-D, A5 = 4 ppm Auxin with 2,4-D, A6 = 5 ppm Auxin with 2,4-D. Observation data obtained were tested using the F test (Anova) and if it showed significant or significantly different results, further, the Smallest Significant Difference (BNT) testing was carried out at the level of 1%. The results of the analysis showed that the treatment of giving variations in the concentration of growth regulating agents auxin 2,4-D in sugarcane callus induction had a very real effect on the smoothy current, callus texture and callus size.
