ABSTRACT



This study aims to obtain a liquid alternative medium for the growth of Rhizobium spp. which has faster growth power compared to selective media (YMB) and also has affordable prices both in terms of financial and in terms of availability in the field. The study was conducted in November 2018 until May 2018. Activities were carried out at the Jember State Polytechnic Biosciences Laboratory. This study uses 4 types of media with 8 different treatments. Each treatment was repeated 5 times to obtain 40 experimental units. The data obtained is then analyzed qualitatively, quantitatively and t-test. Quantitative data analysis was carried out using TPC (Total Plate Count) and qualitative data was obtained through objective observation of bacteria by observing gram staining, observing bacterial growth speed and observing bacterial purity. The results of this study indicate that the conventional medium of bran waste water produced 5.94 x 1024 and was able to multiply bacteria by 40.5% faster and 6 times more financially efficient than the selective media of Yeast Monnitol Agar. The number of population produced between aseptic media and conventional media showed the results of 5.94 x 10-2 with the t test showed the results were not significantly different, so conventional media could substitute aseptic media and the whole sample had fast growing or fast growth types.
