ABSTRAK

Penelitian ini bertujuan untuk Mengkaji pertumbuhan bakteri Rhizobium spp pada media buatan, Mengetahui pengaruh dari masing-masing media perlakuan, Mengetahui pertumbuhan jumlah koloni dari media YEMA, media granul dan media serbuk, Mendapat media tumbuh mikroba yang mampu mendukung pertumbuhan mikroba yang cepat dengan jumlah sel yang tinggi, Mengetahui viabilitas bakteri pada media buatan. Penelitian dilaksanakan bulan Desember 2017 hingga bulan Mei 2018. Kegiatan dilakukan di Laboratorium Biosains Politeknik Negeri Jember. Penelitian ini menggunakan 3 macam media perlakuan dengan 3macam formulasi yang berbeda. Masing-masing perlakuan diulang sebanyak 7 kali sehingga diperoleh 21 unit percobaan. Data yang diperoleh kemudian di lakukan analisis secara kualitatif, kuantitatif dan uji t. Analisis data secara kuantitatif dilakukan dengan menggunakan TPC (Total Plate Count) dan data kualitatif diperoleh melalui pengamatan bakteri secara objektif dengan menggunakan media congo red + YEMA untuk pengamatan kemurnian bakteri, YEMA + Brom Thymol Blueuntukpengamatan kecepatan pertumbuhan bakteri,gram staininguntuk mengetahui jenis gram bakteri.Hasil penelitian ini menunjukan bahwa media formulasi granuldan serbuk dengan daya simpan 3 bulan memiliki jumlah koloni 4,90E+19 CFU/gramdanJumlah populasi yang dihasilkan media formulasi serbuk menunjukkan hasil 5,16E+19 CFU/gram melebihi standar minimum jumlah bakteri yang terkandung dalam pupuk hayati, sehingga layak untuk dijadikan sebagai pudengan uji t test menunjukan hasil tidak berbeda nyata, sehingga media formulasi serbuk dan granul dapat mensubtitusi media YEMAdan keseluruhan sample memiliki tipe pertumbuhan fast growing dengan ciri berwarna kuning jika ditumbuhkan pada media YEMA+BTB (Brom Thymol Blue).

Effication of Rhizobium spp. Bacteria Growth on Edamamae Soybean Waste-Based Media as the Subtitution of Yema Media

ABSTRACT
This research aims to examine the growth of Rhizobium spp bacteria on artificial media, to determine the effect of each treatment media, to know the growth of the number of colonies from YEMA media, granule media and powder media, to obtain microbial growth media that can support rapid microbial growth with cell counts, to know bacterial viability in artificial media. The study was conducted in December 2017 until May 2018. Activities were conducted at the Biosciences Laboratory Politeknik Negeri Jember. This research uses 3 types of treatment media with 3 different types of formulations. Each treatment was repeated 7 times to obtain 21 experimental units. The data obtained is then analyzed qualitatively, quantitatively and through t-test. Quantitative data analysis was conducted by using TPC (Total Plate Count) and qualitative data was obtained through objective bacterial observation using congo red + YEMA media for observing bacterial purity, YEMA + Thymol Blue Bromine for observing the bacterial growth speed, gram staining to determine the type of bacteria gram. The results of this study indicate that the granules and powder formulation media with a shelf life of 3 months had a colonies number of 4.90E + 19 CFU / gram and the population produced by powder formulation media showed the results of 5.16E + 19 CFU / gram exceeding the minimum standard number of bacteria contained in biofertilizers, so that it is feasible to be used as pu by t test showed no significant difference, so that the powder and granule formulation media can substitute YEMA media and the whole sample has a fast growing growth type with yellow characteristics if grown on YEMA + BTB media (Brom Thymol Blue).
