ABSTRACT
The iles-iles plant (Amorphophallus muelleri Blume) is a tuber crop that has the prospect of being developed as an alternative food in Indonesia. The lack of availability of superior seeds and the production of iles-iles tubers is still very limited and the growth of the old hence the need for alternatife multiplication with tissue culture. This study aims to determine the ratio of TDZ and NAA concentrations of the most optimal in the induction of buds iles - iles. Implementation of this research was conducted in September 2017 until February 2018. The place of research was carried out in labolaturium of culture network of State Polytechnic of Jember. This research was conducted by using a complete randomized design factorial with 2 factors, 9 treatments, 3 replications, and each replication there were 4 samples. The first factor is plant growth regulator TDZ consists of three levels namely 0.25, 0.50, 0.75 mg / l. The second factor is plantgrowth regulator NAA 0.10, 0.20, 0.30 mg / l. The data analysis used is Analysis of Variance which then tested by using BNT 5%. The results showed that addition of TDZ with a concentration of 0.75 mg / l had a very significant effect on the shoot height parameters at 14 weeks. In the addition of TDZ with concentration 0.50 mg / l have real effect on parameter of shoot number at age 14 with mean. In addition of NAA with concentration 0.30 mg / l have real effect to the appearance of buds iles iles. Based on these observations it can be seen that the addition of TDZ and NAA Growth Control Substances responds to the growth of iles-iles plants, especially in the formation of buds, buds, shoots, and number of leaf of iles-iles.
